RESUMO:

Sulcatone (SU), 6-methylhept-5-en-2-one, is found in Citrus plants species. It is used as flavoring ingredient and has allelochemical properties. In this study, the inclusion complex of sulcatone (SU) and β-cyclodextrin (β-CD) were prepared (using a spray drier), characterized and analyzed by intermolecular interactions of the complex using computational methods and evaluated sulcatone’s vasorelaxant effect. Evidence of the inclusion complex formation was provided using IR, XRD, TGA, DSC and 1H NMR techniques and further deduced by pharmacological experiments, which showed significant changes in relation to the starting material. The formation constant (337.6 mol L-1) and thermal stability were determined by HS-GC-MS, whose data was similar to that obtained from computational calculations using the ONIOM model (our own n-layered molecular orbital and molecular mechanics). The encapsulation efficiency was 37%. From theoretical calculations, it was possible to evaluate the stability and to determine the mode of orientation of the host (SU) in the cyclodextrin cavity. In normotensive rats, oral administration of the SU-β-CD (100mg/kg, po.) inclusion complex produced a more pronounced hypotensive effect when compared to non-complexed SU. These results demonstrate that the inclusion complex SU-β-CD (100mg/kg, po.) may be used as alternative in the treatment of hypertension.
